Does low oxygen change how plant-derived coumarins affect cells in the laboratory?
The pattern depended on the compound: low-dose galbanic acid showed significant toxicity under low oxygen, while auraptene showed similar toxicity under normal and low oxygen. Umbelliprenin had low but stable toxicity across oxygen conditions and all three measured time points, at the highest concentration tested.
Details
Natural coumarins are described as valuable agents that produce anticancer effects and/or increase sensitivity to therapeutic modalities. Galbanic acid (GBA), auraptene (AUR) and umbelliprenin (UMB) are coumarins derived from Ferula species with various pharmaceutical activities. This in vitro laboratory experiment aimed to compare the toxic effects of these three compounds on human lymphoma cells under normoxia and hypoxia.
GBA and AUR were extracted from the roots of F. szowitsiana, and UMB from the roots of F. persica, all using thin-layer chromatography. MT-2 cells were treated with each compound for three consecutive treatment periods under oxygen concentrations of 21% (normoxia) and 2% (hypoxia). At the end of each treatment, MT-2 cell viability was measured using a resazurin dye-based colorimetric assay.
Low doses of GBA, 10 and 20 µM, produced significant toxic effects under hypoxia (p < 0.0001). AUR at 40 µM showed similar toxicity under normoxia and hypoxia. UMB was the only coumarin to show cytotoxic effects at every measured time point—48, 72 and 96 h—under both normoxia and hypoxia, although its concentration was the highest, at 80 µM.
The authors describe this as the first report indicating that GBA was the most toxic coumarin against ATL cells under hypoxia, that AUR produced similar effects under normoxia and hypoxia, and that UMB's low toxicity remained stable over time and across oxygen concentrations. They recommend future in vitro studies using other ATL cell lines to better evaluate the toxic effects of GBA, AUR and UMB.
